Karyotyping Web Activity KEY The Karyotyping Activity at The Biology Project will help you learn about human karyotyping using digital images of chromosomes from actual human genetic studies. Objective: Complete the web activity to practice completing and analyzing karyotypes. Procedures: 1.

2. Follow the tabs at the bottom of each page to complete the steps for each of the three patients, and follow this paper as a guide and to read both this paper and the website carefully. 3. For EACH patient, do the following: a. Patient A: Read Patient A’s history, and record the following: i.

What or who is the patient? The patient is an almost full-term fetus from a 40-year old woman.

ii.

Where or how were the chromosomes retrieved? The chromosomes were obtained from fetal epithelial tissues through the amniocentesis process.

iii.

Click on “Complete Patient A’s Karyotype”, and complete the karyotype following the prompts.

iv.

When you have completed the karyotype, follow the instructions to interpret the karyotype. Write down the notation for Patient A’s karyotype. (It should be a number— the total number of chromosomes and then XX or XY to show male or female…for example, a normal female would be 46XX) 47, XX, +21

v.

Using the table, write down your diagnosis for Patient A (What disorder to they have?) and why you give this diagnosis. Down’s syndrome, because they have trisomy 21 or an extra #21 chromosome

Click on “Complete Patient B’s Karyotype”, and complete the karyotype following the prompts.

iv.

When you have completed the karyotype, follow the instructions to interpret the karyotype. Write down the notation for Patient B’s karyotype. 47, XXY

v.

Using the table, write down your diagnosis for Patient B (What disorder to they have?) and why do you think that. This patient has Klinefelter's Syndrome because he has an extra “X” sex chromosome.

c. Patient C Read Patient C’s history, and record the following: i. What or who is the patient? Patient C died shortly after birth with many anomalies, including a cleft lip and polydactyly. ii.

Where or how were the chromosomes retrieved? Chromosomes were obtained through a tissue sample.

Click on “Complete Patient C’s Karyotype”, and complete the karyotype following the prompts.

iv.

When you have completed the karyotype, follow the instructions to interpret the karyotype. Write down the notation for Patient C’s karyotype. 47, XY, +13

v.

Using the table, write down your diagnosis for Patient C (What disorder to they have?) and why do you say that. This patient had trisomy 13 syndrome because they had an extra chromosome #13.
